Rakovina Secures Exclusive AI Drug Discovery Profile and Extends Optimization Agreement
Rakovina Therapeutics Inc. (RKV:TSX.V; RKVTF:OTCPK; 7JO0:FSE) announced that it amended and extended its research services agreement with Vancouver-based Variational AI Inc., granting Rakovina exclusivity over the target product profile being pursued for its kt-5000 ATR inhibitor program.
Under the amendment, Variational AI will not design, generate, or deliver to another party compounds intended to meet the particular combination of potency, selectivity, drug-like properties, and brain penetration defined by Rakovina during the agreed term. Variational AI will also conduct additional AI-powered lead optimization cycles beyond the five completed to date, at no additional cost to Rakovina through year-end.
Variational AI will use its Enki generative AI platform to generate and prioritize compound designs in iterative optimization cycles. Rakovina retains control over which candidates advance to laboratory testing and further development. In exchange for the extended term and exclusivity, Variational AI has been granted increased milestone payments upon nomination of a drug candidate and completion of an out-licensing transaction. Payment amounts were not provided in the announcement.
The kt-5000 program consists of small-molecule inhibitors of ATR, a central regulator of the DNA damage response. Rakovina previously reported preclinical data supporting potent ATR inhibition, drug-like properties, and central nervous system penetration in the series. The program focuses on next-generation ATR inhibitors designed to address challenges in treating tumors in the central nervous system.
"The kt-5000 program is targeting a particularly challenging combination of properties: potent and selective ATR inhibition together with the drug-like characteristics and CNS penetration required to reach tumors in the brain," said Dr. Mads Daugaard, Rakovina's President and Chief Scientific Officer, in the company news release. He said exclusivity preserves the differentiation Rakovina is seeking as the program moves toward candidate selection.
"After only five optimization cycles and under 100 compounds synthesized, the kt-5000 work has produced leads we are genuinely excited about," said Handol Kim, Co-Founder and Chief Executive Officer of Variational AI, in the release.
"Using the Enki™ platform, we have advanced kt-5000 in a fraction of the time and at a fraction of the cost of a conventional discovery program," said Kim Oishi, Rakovina's Chief Executive Officer, in the release. Oishi said the additional no-cost optimization cycles extend the collaboration's savings through year-end.
AI Drug Discovery Advances Through Specialized Models and Research Automation
Artificial intelligence had become an increasingly prominent part of pharmaceutical research, with developments in molecular design, therapeutic target identification, and computational modeling demonstrating how specialized systems supported drug discovery. Research efforts had also focused on improving the accuracy of AI-generated predictions and connecting computational analysis with experimental validation.
In its Fall 2026 pharmaceutical AI technology recap, Insilico Medicine described advances in scientific models designed for specific drug discovery tasks, including kinase activity prediction, therapeutic target identification, and the assessment of absorption, distribution, metabolism, excretion, and toxicity. The report stated that specialized models had demonstrated performance matching or exceeding existing benchmarks across more than 70 chemistry and biology tasks.
The report also emphasized the importance of evaluating computational results before incorporating them into pharmaceutical research. Its discussion of AI-assisted antibody development described how models generated ranked predictions and identified candidates for laboratory assessment, while noting that "The scientific output remains a computational recommendation until tested experimentally." The approach illustrated the distinction between AI-generated research findings and experimentally validated results.
According to a September 21 report from McKinsey & Company, investor funding for AI-enabled drug discovery reached US$8.40 billion in 2025, up from US$4.10 billion in 2023, representing a compound annual growth rate of 43%. The report found that selected AI-assisted drug development programs had reached first-in-human or comparable preclinical milestones 15% to 80% faster than large pharmaceutical industry medians, depending on the treatment modality and novelty of the target. McKinsey emphasized that "Novel, causally validated biology remains biopharma's scarcest commodity," identifying biological target validation as a central challenge despite advances in molecular design.
The report also found that molecule design had become the most technologically mature application of AI in drug discovery, supported by structured datasets and rapid experimental feedback. Meanwhile, AI-assisted target identification continued to advance through genomic analysis and specialized models. McKinsey noted that "AI research for target identification is advancing rapidly," while identifying slower laboratory validation and limited biological data as continuing constraints. The analysis distinguished improvements in early research efficiency from clinical outcomes, noting that most AI-originated drug candidates remained in early-stage clinical trials.
An October 14 report from The Stanford Daily examined research into AI-powered biotechnology systems that simulated the structure of a drug discovery organization. The research platform employed tens of thousands of specialized AI agents to identify drug targets, develop molecules, and assess safety and efficacy. Researchers tested the system against more than 50,000 clinical trials, completing the analysis in less than a week.
"We have tens of thousands of AI agents that will look for good drug targets, develop molecules, and also assess the safety and efficacy of those targets," James Zou, an associate Stanford professor of biomedical data science, told The Stanford Daily.
The research also identified a relationship between drug target specificity and clinical outcomes. According to the report, drugs targeting cell-type-specific genes had been 48% more likely to reach the market and had experienced 32% fewer adverse events. Zou emphasized the continued importance of human oversight, stating, "We view the Virtual Biotech as a way to complement and to accelerate human researchers rather than to replace human researchers entirely."
From AI Optimization to Candidate Selection: Research Milestones
Rakovina's May 2026 investor presentation laid out milestones for three cancer drug programs through the first half of 2027. Because the presentation predates the October 14 announcement, its earlier-quarter milestones are historical plans, not confirmation that each activity was completed.
The kt-5000AI program uses Enki to develop dual ATR inhibitors for solid tumors, PTEN-deficient cancers, and central nervous system metastases. The May presentation reported 138 molecules predicted and 43 synthesized, with potent and selective leads confirmed and central nervous system penetration and tolerability observed in vivo. It also described presentations at the Society for Neuro-Oncology meeting in November 2025 and at the American Association for Cancer Research meeting in April 2026.
In its April 2026 AACR poster summary, Rakovina reported generating and synthesizing small molecules designed as brain-penetrant ATR inhibitors that also targeted the mTOR pathway — an alternative pathway that becomes activated upon PTEN loss/mutation. It described varying levels of central nervous system penetration, activity, and metabolic stability, and said Compound D showed equal in vivo efficacy compared with ceralasertib, with decreased weight loss and no signs of hematological toxicity. Further optimization was described as ongoing.
The May presentation listed in vivo model data at peer-reviewed meetings, lead optimization results through an expanded Variational AI collaboration, lead selection, and ongoing partner discussions among the kt-5000AI milestones for 2026. Its quarterly schedule listed iterative in vitro compound testing and refinement in Q2, in vivo ADME and efficacy work, and advancement of lead candidate selection in Q3, and an ATR lead selection in Q4. The October 14 release confirmed additional Enki optimization cycles through the end of the year, but did not say a development candidate had been selected.
For the first half of 2027, the May presentation scheduled Good Laboratory Practice toxicology and investigational new drug-enabling studies for the ATR program.
The second program, kt-3000, comprises dual PARP/HDAC inhibitors directed toward PARP-resistant adult and childhood cancers, including Ewing sarcoma. Rakovina described a joint venture with NanoPalm Ltd. combining its AI drug discovery work with NanoPalm's self-targeting patterned lipid nanoparticle delivery system, with Rakovina retaining intellectual property rights. The presentation reported research published in Clinical Cancer Research and an April 2026 AACR presentation.
The April AACR poster summary described encapsulation of the kt-3283 inhibitor in patterned lipid nanoparticles designed using the EnsaliX AI platform. Rakovina reported uniform particle size and surface charge, stable colloidal behavior, and a hexadome structure. The identified next research steps involved in vitro and in vivo characterization to confirm PARP and HDAC activity, determine ADME properties, and assess efficacy in tumor models.
For kt-3000, the May schedule listed the delivery of NanoPalm's first lipid nanoparticle formulation in Q2 2026, along with in vivo ADME and efficacy testing of formulations. Continued in vivo studies were listed for Q3. Separate 2026 milestones included joint venture funding, an antibody-drug conjugate partnership, and nano-lipid formulation data at peer-reviewed meetings. Good Laboratory Practice, toxicology, and investigational new drug-enabling studies were scheduled for the first half of 2027.
The third program, kt-2000AI, targets brain-penetrant PARP inhibition for breast, ovarian, and prostate cancers. The May presentation described screening billions of compounds through Deep Docking, assessing 389 compounds against PARP, and confirming leads with PARP selectivity and drug-like properties. It also identified a November 2025 Society for Neuro-Oncology presentation covering PARP1/PARP2 assay data, ADME profiling, and pharmacokinetic comparisons against AZD9574 and olaparib.
The May schedule assigned a round of AI output and initiation of in vitro testing for kt-2000AI to Q3 2026. Another round of AI output and advancement of lead candidate selection were scheduled for Q4, followed by PARP program lead selection in the first half of 2027.
At the corporate level, the May milestone schedule identified AI partner strengthening, scientific advisory board talent, an AACR conference, and finalization of a Saudi joint venture among Q3 2026 items. It listed institutional funding and attendance at the SNO, AACR-NCI-EORTC, and BIO Middle East conferences for Q4. The development of strategic pharmaceutical partnerships appeared in its first-half 2027 plans.
